MICROBIOLOGICAL STUDIES OF PHARMACEUTICAL DEVELOPMENT OF GRANULES FOR PHARMACOCORPTION OF MASTROPHY

Fibrocystic mastopathy is one of the most common benign diseases of the mammary glands, characterized by dishormonal and proliferative changes and has a tendency to increase in prevalence. Among modern pathogenetically based approaches to its therapy, phytotherapy occupies an important place, the effectiveness of which is due to the complex action of biologically active substances of plant origin. A promising direction is the creation of phytopreparations in the form of granules, which provide ease of use and optimal pharmacotechnological characteristics. The aim of the work was to study the antimicrobial properties of experimental granules based on phytoextracts, represented by three technological samples, and to justify the choice of the optimal preservative. Microbiological studies of experimental samples of granules based on phytoextracts were carried out using reference test cultures of gram-positive and gram-negative bacteria, as well as yeast-like fungi Candida albicans. Antimicrobial activity was determined by the agar diffusion method with subsequent assessment of the zones of inhibition of the growth of microorganisms. The preparation of microbial suspensions was carried out according to the McFarland standard using 24–72-hour cultures. According to the results of determining the antimicrobial activity, it was found that all the studied granule samples exhibit a weak antibacterial effect on test microorganisms, as evidenced by the diameters of the growth inhibition zones within 12–15 mm. The greatest sensitivity was found for Staphylococcus aureus, Escherichia coli and Bacillus subtilis. At the same time, Sample 3 was characterized by a slightly higher antimicrobial activity against Pseudomonas aeruginosa, Proteus vulgaris and Candida albicans, which was the basis for its selection for further studies. At the next stage, it was found that the sample of granules with sorbic acid at a concentration of 0.1% provides the best antimicrobial profile compared to sodium benzoate and a mixture of nipagin and nipazole. It was established that experimental samples of phytogranules exhibit antimicrobial activity, with the most pronounced indicators characteristic of the Sample 3 of granules. According to the results of the study of the effectiveness of preservatives, it was established that Sample 3 provides the best antimicrobial profile among the studied ones.
